Goal of this project is to test DNA metabarcoding methods for stream assessment. At 20 sample sites kick samples were taken and macroinvertebrates identified based on morphology. Afterwards, DNA from the bulk samples was extracted and the COI gene amplified. Sequencing was done on the illumina system.Main research objectives:- testing of presorting samples by specimen biomass, to reduce sequencing depth (large organisms will contribute substantial amount of DNA in the extraction, thus presorting of specimens and adjusting of used amount of tissue migth help reducing this bias).- Comparison of performance of traditional morphological identification vs. DNA metabarcoding of complete kick samples.
