Our study represents LKB1 defects lead to the loss and dysfunction of adipose tissue ILC2s.And we found that LKB1 regulate ILC2s homeostasis and cytokines function in a cell-intrinsic manner. In high-diet-fed mice,LKB1 in ILC2s protects against metabolic disturbances and fat fibrosis.LKB1 deficient in ILC2 impaired the oxidative phosphorylation and led to mitochondria loss though more mitophagy. mRNA profiles of VAT ILC2s from 8-week old R5/+(WT) and R5/+Stk11f/f(KO) male mice
本研究证实,LKB1缺陷会导致脂肪组织2型固有淋巴细胞(ILC2s,innate lymphoid cell type 2)的丢失与功能异常。我们发现,LKB1以细胞自主性方式调控ILC2s的稳态及细胞因子功能。在高脂饮食饲喂的小鼠中,ILC2s中的LKB1可抵御代谢紊乱与脂肪纤维化。ILC2中LKB1缺失会损伤氧化磷酸化过程,并通过增强线粒体自噬(mitophagy)导致线粒体丢失。本研究的测序数据来自8周龄雄性R5/+(野生型,WT,wild type)与R5/+Stk11f/f(基因敲除型,KO,knockout)小鼠的内脏脂肪组织(visceral adipose tissue, VAT)ILC2s的mRNA转录组谱。
