Cells were incubated with an ATP-regenerating
system, Ran-GTP, NTF2, and an NLS-TD-Tomato cargo in the presence and absence
of Kap1- α and Kap1-b. Amount of NLS-TD-Tomato accumulated in the nucleus was measured by quantifying
the average fluorescence signal in the nucleus, as defined via Hoechst
staining.
