We performed RNA-seq analysis for organs (ovary, testis, liver, muscle, kidney, heart, gill, eye, and brain) of males and females in the wild-type fish of the Cab strain, respectively, to construct the sex or organ-specific gene expression profile. This profile could provide the expression level of a gene of interest and could support a variety of genetic analyses. Especially in the last decade, genome editing technology has developed remarkably, facilitating in-frame knock-in, such as fusion of a reporter gene to the open reading frame of interest. We have shown using the RNA-seq data that the expression patterns of green fluorescent protein in GFP knock-in medaka mimicked those of the endogenous gene. Thus, our expression profiles, in combination with genetic modification techniques such as genome editing, will allow us to analyze gene function in this species from multiple perspectives.
