Vibrio alginolyticus is a marine opportunistic pathogenic bacterium that can infect a variety of aquatic animals. In this study, V. alginolyticus H1 degraded collagen extracted from the skin of cuttlefish Sepia pharaonis. The catalytic domain of ValC of ColA from V. alginolyticus H1 was cloned, and the recombinant ValC (rValC) was purified using Ni-NTA agarose, and its catalytic activity and expression were characterized. To further analyze the function of ColA, a colA gene knockout strain was first constructed using sucided plasmid pDM4. Cuttlefish S. pharaonis were separately challenged with wild type strain H1 and , and the livers were collected for the transcriptomic analysis.
