Gene expression of cardiomyocytes isolated from vehicle and Doxorubicin challenged mice

The goal of this study was to identify differentially expressed genes in cardiomyocytes isolated from vehicle and Doxorubicin treated mice MX1gfp mice (Jackson Laboratory strain 033219, n = 3 per group) were intraperitoneally (i.p.) injected with vehicle (PBS) or one dose of 25 mg/kg of Doxorubicin. Forty eight hours (48 h) post injection, hearts were perfused, digested, and cardiomyocytes were isolated. Total RNA was isolated from cardiomyocytes using the Direct-zol RNA miniprep kit (Zymo Research) and subjected to library preparation and RNA sequencing at the Experimental Genomics Core of the Texas A&M Institute for Genome Sciences and Society. FASTQ files were normalized, processed, and analyzed using BaseSpace Sequence Hub (Illumina). The STAR algorithm of RNAseq Alignment V2.0.2 software was utilized to align the results to the Mus musculus/mm10 (RefSeq) reference genome, then RNA-seq Differential Expression V1.0.1 software was used to obtain differential gene expression values and determine statistically significant changes in gene expression via pairwise comparison of experimental groups.
