This dataset contains RNA sequencing data generated from liver tissues of wild-type (WT) and PPARA-null (KO) mice treated with the potent PPARA agonist Wy-14,643 or a control vehicle. The study aimed to investigate the genome-wide transcriptional regulation mediated by PPARA activation and its role in the context of acesulfame-K (AK)-induced non-alcoholic fatty liver disease (NAFLD). Liver samples were collected from four experimental groups: WT control, WT treated with Wy-14,643, KO control, and KO treated with Wy-14,643. Comparative transcriptome analysis identified key differentially expressed genes and pathways downstream of PPARA, providing insights into its dual role in regulating hepatic phospholipase C beta (PLCB) and diacylglycerol kinase eta (DGKH) within the sweet taste receptor (STR) signaling pathway. This data is crucial for understanding the molecular mechanisms by which PPARA modulates hepatic lipid and glucose metabolism upon artificial sweetener exposure.
