以林业锥栗总苞为原料进行热水浸提,经过醇沉、脱色素、脱蛋白以及DEAE-Sepharose Fast Flow离子交换柱和Sephadex-G50凝胶柱的分离纯化得到一种阿拉伯半乳聚糖组分(CHIP3),再对该组分进行理化性质及抗氧化活性分析。结果表明:SJP-2总糖含量为 98.01%,不含蛋白,其纯度为99.61%,是一个低分散度,分子量较为均一的多糖,分子量为2.44×104g/mol;HPLC分析得知CHIP3为一种杂多糖,其单糖组成及比例为甘露糖(10.7%):鼠李糖(8.7%):半乳糖醛酸(38.21%):半乳糖(13.75%):阿拉伯糖(28.63%);核磁共振及甲基化结果显示该组分主链为1→4链接的半乳糖醛酸和1→2链接的鼠李糖。

Using forestry-harvested Chinese chestnut (Castanea henryi) involucres as raw material, hot water extraction was performed, followed by alcohol precipitation, decolorization, deproteinization, and sequential separation and purification using DEAE-Sepharose Fast Flow ion exchange column and Sephadex-G50 gel filtration column to obtain an arabinogalactan fraction designated as CHIP3. Subsequently, the physicochemical properties and antioxidant activity of this fraction were analyzed. The results demonstrated that: SJP-2 had a total sugar content of 98.01%, was protein-free, with a purity of 99.61%. It was a polysaccharide with low dispersity and relatively homogeneous molecular weight, with a molecular mass of 2.44×10⁴ g/mol. HPLC analysis revealed that CHIP3 was a heteropolysaccharide, with its monosaccharide composition and molar ratio as follows: mannose (10.7%), rhamnose (8.7%), galacturonic acid (38.21%), galactose (13.75%), and arabinose (28.63%). Nuclear magnetic resonance (NMR) spectroscopy and methylation analysis showed that the main chain of this fraction consisted of 1→4-linked galacturonic acid residues and 1→2-linked rhamnose residues.
