The aim of this project was to investigate the role of the mitochondrial protein Tmbim5 in the regulation of mitochondrial calcium (Ca2+) homeostasis and mitochondrial function. Mitochondrial Ca2+ transport plays a key role in cellular energy metabolism, regulation of oxidative phosphorylation, and intracellular calcium signaling. However, the molecular mechanisms controlling mitochondrial Ca2+ levels are still not fully understood.To address this question, the project used the zebrafish (Danio rerio) as an in vivo model. A combination of experimental approaches was applied, including phenotypic characterization of mutant lines, behavioral and morphological analyses, molecular and biochemical assays, measurements of Ca2+ transport in isolated mitochondria, and live imaging of mitochondrial Ca2+ level using genetically encoded fluorescent probes.The results helped clarify the role of Tmbim5 in mitochondrial function and revealed its functional interaction with other components of mitochondrial Ca2+ transport pathways, particularly the Na+/Ca2+ exchanger NCLX (Slc8b1) and the mitochondrial Ca2+ uniporter (MCU).
