Barley mutants with lesions at the lys3 locus show larger than normal sized embryos which show an increase in lysine levels and reduced starch content, resulting in improved grain nutritional value. So far, four independent lys3 alleles have been identified in barley: Risø18, Risø19, Risø1508 in the cultivar Bomi and M1460 in the cultivar Minerva. The lys3 locus has been mapped previously to chromosome 5H at approximately 42 cM based on linkage information present in Barley Genetic Stocks descriptions for barley genes. The aim of this study was to discover the genetic background of the lys3 mutation.Grains used in this study were acquired from publically-available germplasm collections: BBSRC Cereals Collection, JIC (Bomi, Risø1508), or were a kind gift of Birthe Møller Jespersen, University of Copenhagen, Denmark (Minerva, Risø18, Risø19, and M1460).
