Bulk RNA sequencing of kidney in newborn non-CF and CF pigs

We did bulk RNA sequencing in newborn cystic fibrosis (CF) and non-CF pig kidney. We compared kidney gene expression profiling between non-CF and CF pigs. RNA sequencing results showed that there is not significant difference between non-CF and CF in terms of gene expression, suggesting that CFTR knockout does not affect kidney development in newborn pigs. Overall design: Kidney cortex tissue were collected from 6 wild-type (WT), 6 CFTR+/- (HET), and 11 CFTR-/- (CF) newborn pigs. We tried the best to collect the tissue from a similar position of the kidneys. RNA was extracted using a RNeasy Lipid Tissue Kit (Qiagen). cDNA libraries were prepared using the Illumina TruSeq Stranded Total RNA with RiboZero, followed by sequencing on NovaSeq 6000 (Illumina).
