Dietary isoleucine is a significant determinant of metabolic health and longevity. In this study, we found that even at an advanced age, both male and female mice received significant metabolic benefits from a low isoleucine diet. We collected liver tissues for transcriptomic analysis and the results found that the majority of age-driven changes is ameliorated with the administration of low isoleucine diet. Overall design: In this transcriptomic study, we treated 20-month-old C57BL/6J.Nia mice of both sexes with either a control or a low isoleucine diet, and 6-month-old C57BL/6J.Nia mice with a control diet. They ate their respective diets for 4 months before being sacrificed after a overnight fast (16 hr) and refeeding (3 hr). The liver was flash frozen in L2N and the RNA extracted.
