To establish a severe steatotic model, eight-week-old rats (200-220 g) were fed a high-fat diet (China National Institutes for Food and Drug Control), which consisted of 15% (w/w) triglyceride, 15% (w/w) sucrose, 10% (w/w) egg yolk powder, 1% (w/w) cholesterol, 0.2% (w/w) bile salt, and 58.8% (w/w) basic feed, for a total of 20 weeks. To induce ischemia-reperfusion injury (IRI), noninvasive arterial clips were used to block the portal vein and hepatic artery supplying the left and median lobes of the liver under general anesthesia and removed after 80 minutes of sustained ischemia. One day before surgery and immediately after reperfusion, 2 x 10^6 HO-1/BMMSCs were injected via the penile dorsal vein or portal vein. Rats were euthanized 24 hours after reperfusion, and 0.2 g fresh rat fecal samples were collected and stored in airtight sterile freezing tubes in liquid nitrogen for 16S rDNA amplicon sequencing.
