We assessd the transcriptional profile of T. rubrum during grown on different protein sources to mimic the natural infection sites (keratin or elastin) and exposed to trans-chalcone in order to clarify the mechanisms involved in trans-chalcone antifungal activity About 1.6 10^6 conidia was added in 20mL of liquid Sabouraud followed by an incubation for 72h at 28ºC under shaking at 150 rpm. Then the resulting mycelia was transferred to six different conditions:i) Minimal medium Cove's with nitrate and glucose, ii) Keratin medium: Cove’s minimal medium with 0.5% bovine keratin powder; iii) Elastin medium: Cove’s medium with 0.25% elastin; iv) Minimal medium added of 0.24µg/mL of trans-chalcone and nitrate and glucose; v) Minimal medium with 0.5 % of keratin, added of 0.24µg/mL of trans-chalcone and vi) Minimal medium with 0.25% of elastin, added of 0.24µg/mL of trans-chalcone. At least two independent replicates were performed for each condition
