We established a platform for the brain organoid culture by using human decellularized brain extracellular matrix (BEM) and a microfluidic device. This engineering concept of reconstituting brain-mimetic microenvironments facilitates development of a reliable culture platform for brain organoids, enabling effective modeling and drug development. Overall design: Matrigel-encapsulated brain organoid vs BEM-encapsulated brain organoid and conventional plate culture encapsulated with BEM vs microfluidic culture encapsulated with BEM.
