This study provides a systematic comparison and comprehensive evaluation of current commercially available single-cell RNA sequencing (scRNA-seq) technologies. A single peripheral blood mononuclear cell (PBMC) sample was used to illustrate key features and performance metrics of these technologies. Technologies were evaluated and ranked based on multiple categories such as kit performance, sample type compatibility, cost-effectiveness, and experimental duration. One of the most prominent findings was the superior performance demonstrated by the 10X Fixed RNA Profiling kit which uniquely features transcript detection through probe hybridization as well as extensive sample compatibility. However, cost and protocol duration are crucial determinants in kit selection with Fluent Biosciences and Singleron proving to be the most cost-effective, and 10X Genomics offering the most time efficient protocols. This work not only helps to inform decision-making for researchers seeking a suitable scRNA-seq technology but also contributes a rich dataset of over 218,154 annotated cells to the field, aiding in further technology and algorithm development.
