Gene expression profile at single cell level of murine B cells from different compartments of naive young adult mice

In the context of understanding thymic T cell selection facilitated by local B cells, we sought to compare thymic B cells with their counterparts from other compartments in naive young adult mice. B cells from spleen, inguinal lymph nodes, bone marrow, thymus and blood (each pooled from two mice) were isolated by fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq (10x).为阐明局部B细胞介导的胸腺T细胞选择机制，本研究旨在对比未感染年轻成年小鼠体内，胸腺B细胞与其他免疫隔室来源的同源B细胞。我们从脾脏、腹股沟淋巴结、骨髓、胸腺及血液中分离B细胞（每组样本均取自2只小鼠的混合细胞），通过荧光激活细胞分选（fluorescence-activated cell sorting, FACS）完成分离，并采用10x平台开展单细胞RNA测序（single-cell RNA sequencing, scRNAseq）分析。
