This dataset accompanies the manuscript: "Lymphoma Fingerprints in Healthy Individuals Uncovered through Single-Cell DNA Methylation Analyses." It includes raw and processed data from single-cell multiomic profiling of human B cells, integrating single-cell DNA methylation (scDNAm) and surface-marker expression. Contents Raw Data: scDNAm matrices (per sample): raw methylation counts from the scTAM-seq pipeline. Surface-marker data: raw antibody-derived tag (ADT) counts per cell. EM-seq: count matrix per sample. scRNA-seq: Celescope outs folder per sample/run. Processed Data: Final multiomic Seurat object (RDS format), containing three assays: DNAm.counts: raw DNA methylation values DNAm: binarized DNA methylation data ADT: antibody-derived tag counts matrix Seurat object with scRNA-seq data EM-seq final counts matrix: % of methylated reads per CpG per sample. Metadata Integrated in the Seurat object and includes: QC metrics Cell-type annotations B-cell receptor (BCR) annotation and related features. Sample and donor metadata. Samples scTAM-seq: 12 samples derived from 9 healthy donors, from peripheral blood and bone marrow. Two samples (T23 and T49) are undigested controls (no HhaI enzyme) and can be used to assess false-negative rates. scRNAseq: 4 samples of CD19+ B cells from blood. Two different populations (CD5high and CD5low) were sorted from each sample, T48 and T49. EM-seq: 6 samples in total from 3 donors. For each donor, we have an enriched (S1, S3, S5) and a control sample (S2, S4, S6). All donor and sample-specific information is available in the Seurat object metadata and detailed in the Supplementary Material of the published article.
