The Washington University School of Medicine (WashU), in collaboration with the Baylor College of Medicine and the University of California at San Francisco, constructed a physical map of the Ajellomyces capsulatus strain G217B genome using a fosmid fingerprint-based approach. They generated three independent restriction endonuclease fingerprints for each analyzed clone from a fosmid library representing approximately 10X genome coverage. The data were combined and used in FPC for automatic and manual contig assembly. WashU anchored the physical map to their WGS assembly using fosmid paired-end sequences, which resulted in the joining of existing physical map contigs into 84 clusters containing 9,551 fosmid clones.
