Single cell transcriptome of the mouse larynx

Microbiota play a critical role in training and development of major components of host innate and adaptive immune system. We present the cellular landscape of the upper airway, specifically the larynx, by establishing a reference single-cell atlas and dissected the role of microbiota in laryngeal cell development and functions at a single-cell resolution by comparing the transcriptomes of conventionally raised and germ-free mouse larynges. Single cell suspension of the mouse larynx was prepared for conventionally raised and germ-free mice and sequenced and analyzed following 10x genomics pipeline.
微生物群(microbiota)在宿主先天免疫与适应性免疫系统核心组分的驯化与发育过程中发挥关键作用。本研究通过构建参考单细胞图谱,呈现了上呼吸道(特指喉部)的细胞全貌,并通过对比常规饲养与无菌(germ-free)小鼠的喉部转录组,以单细胞分辨率解析了微生物群在喉部细胞发育与功能中的调控作用。针对常规饲养与无菌小鼠,研究人员制备了其喉部的单细胞悬液,并依照10x基因组学(10x Genomics)测序流程完成了测序与数据分析。
