The study aims at understanding open chromatin regions in ovaries of blood-fed females (24h and 48 h post-blood feeding) and in early embryos (3 h and 5 h post-oviposition) of the yellow fever mosquito Aedes aegypti. Ready for sequencing libraries were prepared using ATAC-Seq kit from Active Motif (Cat No. 53150) following the manufacturer's recommendation. For 3 h- and 5 h-old embryo samples, ~150 eggs were homogenized in 400 µL ATAC Lysis Buffer. For ovary samples 24 h and 48 h post-blood meal, ovaries from 50 females were dissected and immersed in ice-cold PBS before homogenizing in 400 µL ATAC Lysis Buffer. An aliquot of approximately 750,000 cells were used for tagmentation step, which was followed by 11 cycles of PCR amplification. The libraries were sequenced using Illumina NovaSeq6000 platform.
