Endothelial cells (ECs) can detect oxygen and nutrient shortages in tissues and are able to adapt by inducing proliferation and migration into ischemic areas. Here, we present results that elucidate the striking role of a novel, EC/mural cell-specific gene, Apold1, in regulating pathological angiogenesis in ischemia. Screening of the transcriptomic profile by bulk RNAseq of wild type (WT) and Apold1-/- isolated muscle endothelial cells (ECs) revealed minor transcriptional differences upon Apold1 knockout. In contrast RNA sequencing on primary ECs derived from Apold1-/- or WT ECs, which were briefly (16 hrs) cultured in angiogenic medium, showed striking differences. Comparative mRNA gene expression analysis of ECs that were isolated by FACS sorting from mouse hind leg muscles. Two experiments were performed, one (5vs5) comparing Apold1KO vs WT in sorted cells directy after sorting, and one experiment (3vs3) comparing Apold1KO vs WT in sorted cells after briefly (16 hrs) cultureing in angiogenic medium
