ight sheet microscopy imaging dataset of CAR-T-cell-mediated cytotoxicity

Research on CAR-T cell states is crucial for understanding immunotherapy mechanisms. Previous studies in live cells have been primarily limited by phototoxicity, resolution, and throughput，making it difficult to conduct further research and observation on cell states. To enable more detailed studies of cell states, we have constructed a microscopy imaging system that enables subcellular resolution, low phototoxicity, high imaging throughput, and data reconstructing automation. Using this system, we have generated and shared over 500 image sets that capture the cytotoxic processes of CAR-T cells against tumor cells. The data provide an isotropic spatial resolution of 320nm, a temporal resolution of up to 2.5 seconds per volume, and long-term observation spanning up to 5 hours. Our present study reported an imaging system, with subcellular resolution, low phototoxicity, high throughput, and data reconstructing automation capabilities, filled an essential gap in the field and offered valuable insights into the cytotoxic processes of CAR-T cells against tumor cells, advanced research in this area obviously.
