Vibrio parahaemolyticus RIMD2210633 (wild-type) was cultured in 2.5 percent (w/v) heart infusion (HI) broth (BD Biosciences, USA), supplemented with either 0 or 71.68 milligrams of ammonium chloride (NH4Cl) per liter, or 6.25 millimoles of sodium taurocholate (TCA) per liter, at a temperature of 37 centigrade with shaking at 200 rpm. The VPA1682 gene mutant was also grown under the same conditions in HI broth. Bacterial cells were harvested during the mid-log phase of growth. Total RNA was extracted, and mRNA enrichment was performed using an Illumina/Ribo-Zer rRNA Removal Kit designed for bacteria. Construction and sequencing of the cDNA library were conducted on an Illumina HiSeq platform.
