We collected fresh fruit body distributed in Neijiang City, Sichuan Province , rinsing off soil and other impurities adhering to its surface with distilled water, and cutting into pieces. The tissue blocks were weighed, ground into powder by adding liquid nitrogen, and treated with 2.0% CTAB for whole genomic DNA extraction. A sequencing library with an average length of 300-500 bp was constructed according to the PE 150 mode of Illumina NovaSeq 6000 platform and loaded for detection. The quality controled data obtained after sequencing was used for genome assembly. SPAdes (v3.15.4) software was used for genome assembly, with the genome sequences of Termitornyces sp. cryptogamus, Termitornyces sp. MG145, Termitornyces sp. JCM 13351 and Termitornyces sp. J132 as reference, according to the isolate mode. Then, the correct, scaffold, and patch function modules in RagTag (v2.1.0) software were used in turn to correct errors, build Scaffolds, and fill gaps in the T. albuminosus (Berk.) Heim assembled genomic sequences. The assembled sequences were further binned and de-redundant using MetaBAT (v2.15) and Redundans (v0.14a) softwares.
