The overarching objective of this project is focusing on the creation of foundational data to establish Duplex Sequencing (DS), an error-corrected next-generation sequencing technology, as a regulatory assay for in vivo mutagenicity testing. DS was applied using a 48 kb panel of endogenous loci spread over the various mouse chromosomes to determine chemically induced mutagenicity. Mouse males were exposed to increasing doses and durations of benzo[b]fluoranthene, a polycyclic aromatic hydrocarbon, and we extracted the DNA of somatic tissues.
