iPSC derived microglial precursors were transplanted into the brains of neonatal mice, FACS sorted out based on GFP+ following 2 months, and compared to in vitro differentiated iPSC derived microglia from the same precursors. Overall design: Microglial Precursors were either transplanted into neonatal mice, plated for in vitro differentiation, or FACS sorted for RNA collection. All samples in the study came from the same batch of differentiated iPSCs of cell line 657-GFP. Transplanted GFP+ cells and in vitro differentiated cells were collected for FACS sorting and RNA extraction following 10 days and 60 days, resulting in a total of 5 sample groups, n=6 for each group. For mice, n=one mouse. For in vitro differentiated microglia, n=3 pooled well of a 6-well plate. For precursors, n=floating cells from two pooled t75 flasks.
