Somatic hypermutation of gene edited heavy-chain antibodies at the IGHG1 CH1-H intron

Raji B cells were gene edited to insert a heavy-chain antibody expression cassette derived from VHH-J3 or -A6, or a control GFP expression cassette. Cells were cultured for 6 months with intermittent sampling. Somatic hypermutation of the inserted cassettes was measured by targeted amplicon sequencing.
