To define the molecular underpinning of LUAD bone metastasis, we performed high-throughput transcriptome sequencing (RNA-seq) using total RNA from LuM and BoM cell subpopulations. Afterwards, differentially expressed genes were used for further analysis. Overall design: We established a mouse model for multi-organ metastases of A549 cells, which generated four bone-metastatic subpopulations (A549-BoM-1, 2, 3, and 4) and lung-metastatic subpopulations (A549-LuM-1, 2, and 3). All of them were used for RNA-seq, and the LuM group was used as negative control.
