This dataset contains archival microsatellite genotypes of the Czech Gold Speckled Hen (Gallus gallus domesticus), a traditional autochthonous chicken breed of the Czech Republic, generated from blood samples collected in 2004–2005. Twelve individuals were genotyped at eight polymorphic microsatellite loci (ADL102, MCW1, MCW5, ADL176, ADL267, ADL158, ADL172, and ADL210) using multiplex PCR and capillary electrophoresis. Allele sizes were scored using GeneScan® and Genotyper® software and manually verified. The dataset provides individual multilocus genotypes and allele frequencies and represents a rare historical molecular baseline for assessing genetic diversity, inbreeding, and long-term genetic changes in Czech poultry breeds.
