ADSC gene expression tendency depends on the medium which was used during cultivation

The effect of culture media to the cultivated cells are not well understood. We evaluated to visualize the medium effects, related with signal pathways and gene expression tendencies by comprehensive gene analysis. The microarray analysis was conducted by using RNAs extracted from adipose derived stem cells in five independent media sets. Four types of xenofree MSC media were selected and used to cultivate ADSCs. These names of media were randomly anonymized and labeled as medium A, B, C and D in the quantitative results, to compare the other cell potency assay results.The actual names of media are listed without particular order; StemPro MSC SFM Xenofree Medium (Gibco/Thermo Fisher), KBM ADSC-4 (Kohjin-Bio), MesenCult ACF Plus culture Kit (STEMCELL technologies) and Cellartis MSC Xeno-Free Culture Medium (Takara Bio Europe AB). A growth-factor containing MSC medium, MesenPRO (MP), was also used to compare with these xenofree media. After cultivation with these media, cells were harvested and total RNAs were extracted from the cells. MSCGM condition(Sample 10 and 11)was a candidate condition as a positive control in this study, but we desided not to use this condition after evaluation because of its high variability in the tube formation assay results.
