To gain more insight into the cellular pathways, whole transcriptome analysis were performed to compare the differentially expressed genes between control and conditional smooth muscle cell specific SDF-1/CXCL12 mutant hearts. Using an optimized data analysis workflow, we mapped about 25 million sequence reads per sample to the mouse genome (build mm10) and identified 15591 transcripts in the hearts of WT and SM-SDF-1 -/- mice. We have detected 143 differentially expressed genes or DEG (more than 1.5 fold, p<0.05), whereas 93 genes were significantly upregulated and 50 genes were significantly downregulated. Further we validated transcriptome data of DEGs with qPCR analysis confirmed the upregulation of genes such as COL8A1, VCAN, BGN and downregulation of RETNLA (FIZZ-1), CXCL14 in KO hearts.
