scRNA-seq analysis of SHH medulloblastomas in Gfap-Cre/SmoM2 mice treated with CT-179 or Pox-resiquimod

Our parallel studies have shown that the OLIG2 inhibitor CT-179 and the TLR7/8 agonist resiquimod delivered in polyoxazoline nanoparticle (Pox-resiquimod) produce significant anti tumor effects when administered to mice with SHH medulloblastoma. To gain insight into the cellular events set in motion by each treatment, we compared sets of replicate mice with SHH medulloblastomas treated with either CT-179 or POx-resiquimod to a common set of untreated control mice with SHH medulloblastoma. We generated replicate mice that developed medulloblastoma by breeding hGfap-Cre and SmoM2 mouse lines to generate Gfap-Cre/SmoM2 mouse pups. We then treated one group of 4 replicates with CT-179 and another group of 3 replicates wih POx-resiquimod, and another 4 replicates served as untreated controls. All mice were harvested at P15 and tumros were dissociated and subjected to scRNA-seq using the Split-Seq-based Parse 1.0 WT kit (Parse Biosciences) and then sequenced on an Illumina S2 flow cell.
