Gene expression in CD30+ L540 cell line induced by different anti-CD30 antibodies that bind to different epitopes

CD30 receptor (TNFRSF8) is highly expressed in some types of lymphoma. Therefore, CD30 is successfully used as a target for antibody-drug conjugate to treat CD30+ lymphoma. In this study, we compared gene expression profiles induced in CD30+ L540 cell line by treating CD30 natural ligand or anti-CD30 antibodies against different epitopes. 12 samples were analyzed. CD30+ Hodgkin’s lymphoma L540 cells were precultured for 16 hrs. They were subsequently treated in the used medium with CD153(CD30L)-Fc fusion protein or anti-CD30 T104, T428, T427 or PW30 monoclonal antibodies for 8 hrs (not 24 hrs=the sample labels were wrong). Biological duplicates were prepared for each treatment. Biotin-labeled cRNA were prepared from the total cellular RNA and subjected to hybridization with the HumanHT-12 v3 Expression BeadChip (Illumina). Data were analyzed using BeadStudio.
