single-nucleus transcriptomic analysis of cortical tissue from control and Dot1l HET 10 week old mice.

We aimed to understand the transcriptional impact of monoallelic loss of DOT1L in cortical nuclei from 10 week old mice. Control and DOT1L HET cortical nuclei were isolated from 10-week-old mice. DOT1L HET mice were heterozygous for floxed DOT1L and both Controls and DOT1L HET were heterozygous for CMV-cre (Strain #:006054). Cortical nuclei were isolated and prepared using the Chromium Next GEM Single Cell Multiome ATAC + Gene Expression reagents. (n = 6 biological replicates combined per sample/genotype, 3 male mice and 3 female mice)

本研究旨在探究10周龄小鼠皮层细胞核中DOT1L单等位基因缺失所产生的转录影响。我们从10周龄小鼠体内分离得到对照组与DOT1L杂合缺失(DOT1L HET)小鼠的皮层细胞核。DOT1L HET小鼠为DOT1L基因侧翼携带loxP位点的杂合子,且对照组与DOT1L HET小鼠均携带巨细胞病毒启动子驱动的Cre重组酶(CMV-cre),品系编号为006054。皮层细胞核的分离与制备采用了Chromium Next GEM单细胞多组学ATAC+基因表达试剂盒(Chromium Next GEM Single Cell Multiome ATAC + Gene Expression reagents)。(每个样本/基因型的生物学重复数为6,包含3只雄性小鼠与3只雌性小鼠)
