Species-specific detection of polar cod (Boreogadus saida) and saffron cod (Eleginus gracilis) eDNA in the Bering and Chukchi seas during the MR21-05C research cruise

To provide an overview of the distribution of polar cod (Boreogadus saida) and saffron cod (Eleginus gracilis) in the Bering and Chukchi Seas, eDNA samples collected during the MR21-05C cruise, which conducted by the 'R/V Mirai' of Japan Agency for Marine-Earth Science and Technology, were analyzed. eDNA samples were collected by filtering surface seawater and seawater obtained from the middle or bottom layers. Filtration was performed using a cartridge filter (Sterivex HV, 0.45 m pore size; Millipore, Burlington, MA, USA). The DNA of both species was quantified in environmental DNA (eDNA) samples utilizing a species-specific TaqMan qPCR assay.为概述北极鳕鱼（Boreogadus saida）与细身宽突鳕（Eleginus gracilis）在白令海及楚科奇海的分布情况，本研究对日本海洋地球科技局（Japan Agency for Marine-Earth Science and Technology）所属“未来号”（R/V Mirai）海洋调查船在MR21-05C航次中采集的环境DNA（environmental DNA, eDNA）样本开展了分析。环境DNA样本通过过滤表层海水及中下层海水采集获得；过滤操作采用筒式过滤器（cartridge filter），具体型号为Sterivex HV型（孔径0.45 μm；密理博（Millipore），美国马萨诸塞州伯灵顿市）。本研究利用物种特异性TaqMan实时荧光定量PCR（TaqMan qPCR）检测体系，对环境DNA样本中两种鱼类的DNA进行了定量分析。
