RNA sequencing was performed on three biological replicates of untransduced (UTD) T cells, CART19 (TNFR2 wildtype) and TNFR2 knockout CART19 produced from normal donors. Overall design: T cells from normal donors were transduced with a replication-incompetent lentiviral vector expressing a second-generation chimeric antigen receptor (CAR) consisting of an anti-CD19 single chain variable fragment (FMC63) fused to 4-1BB and CD3ζ intracellular domains as previously described. TNFR2 knockout CART19 were also transduced with TNFR2 CRISPR Guide RNA in a pLentiCRISPR v2 backbone as previously described. RNA was isolated on Day 8 of production.
