To investigate the transcriptome that might be dysregulated by MYC hyperactivation specifically when DIS3L is downregulated. MYC was induced for 24 hours by adding 60nM 4-OHT with and without DIS3L depletion in human mammary epithelial cells and rRNA-depleted RNA was sequenced. Total RNA (1ug/sample) was used as input for the NEBNext® rRNA Depletion Kit (Human/Mouse/Rat) (NEB). After rRNA depletion, libraries were made using the NEBNext® Ultra™ Directional RNA Library Prep Kit for Illumina®, quantified using KAPA Library Quantification Kit, and sequenced on the NextSeq 500/550. After demultiplexing, reads were processed using SAMtools (v1.4) and aligned using Hisat2 (v2.0.4) with default parameters. The reference genome sequences (hg38/GRCh38) and gene annotation files were obtained from the UCSC Genome Browser.
