Thyroid hormone-responsive genes in primary cultures of rat hepatic cells

cDNA microarray analysis was applied to primary cultures of rat hepatic cells treated with triiodothyronine (T3) at 10-9 M for 24 hrs to identify the differentially expressed genes. A limited number of genes were listed, and only 3 of them, pyridoxal kinase (Pdxk), phosphoe-nolpyruvate carboxykinase 1 (Pck1), and solute carrier family 17 member 2 (Slc17a2), were con-firmed to be up-regulated by quantitative RT-PCR.
为鉴定差异表达基因,本研究采用cDNA微阵列分析(cDNA microarray)对经10⁻⁹ M浓度三碘甲状腺原氨酸(T3)孵育24小时的大鼠原代肝细胞培养物进行分析。本研究仅列出了少量差异基因,其中仅吡哆醇激酶(Pdxk)、磷酸烯醇式丙酮酸羧激酶1(Pck1)及溶质载体家族17成员2(Slc17a2)这3个基因经定量RT-PCR(quantitative RT-PCR)验证存在表达上调。
