In these mock communities, the expected concentrations of 16S rRNA gene were 10<sup>6</sup> copies/μL equally, while the expected concentrations of ITS were 10<sup>6</sup>, 10<sup>5</sup>, 10<sup>4</sup>, 10<sup>3</sup>, 10<sup>2</sup> and 10 copies/μL respectively. In other words, the expected F:B gene ratios of six mock communities were 1:1, 1:10, 1:100, 1:1000, 1:10000, and 1:100000 respectively. The concentrations of 16S rRNA gene and ITS were quantified by SYBR Green qPCR, TaqMan qPCR, SYBR Green ddPCR and TaqMan ddPCR respectively.
