Identification of IKAROS target genes in resting naive CD4 T cells

To identify IKAROS target genes in resting CD4 naive T cells, Naive CD4 T cells were purified from PBMC from healthy donors with EasySep Human Naive CD4 T cell Enrichment Kit (STEMCELL Technologies, Cat 19555).one million of cells were transfected with control siRNA (Dharmacon D-001810-10-05) or IKZF1 siRNA smart pool (Dharmacon L-019092-02) by using P3 Primary cell 4D-Nucleofector Kit and the Lonza 4D-Nucleofector System (Lonza). Cells were cultured in in RPMI-1640 media supplemented with 10% fetal bovine serum (FBS) for 48h until collection. Libraries were prepared using the NuGEN Ovation Whole Blood Kit according to the instructions and sequenced on an Illumina 2500 HiSeq.
