The goal of this set is to build expression data using FACS isolated Ret-positiveNaV1.8 cells harvested from RetFloxedEGFP;NaV1.8Cre embryos (embryonic day E18.5 male or female). These can be used as a time point in comparative analysis of ontogeny of genes expressed at various developmental time points in Ret nociceptors. RNA was extracted from E18.5 FACS isolated dorsal root ganglia dissected from RetFloxedEGFP:NaV1.8Cre mice (EGFP conditionally expressed in Ret population that express Cre from the NaV1.8 locus) and sequenced using Illumina Hiseq 2500 platform. 本数据集的构建目标为:从携带有Ret(Rearranged during transfection)位点loxP侧翼增强型绿色荧光蛋白(EGFP)且NaV1.8基因座驱动Cre重组酶的(RetFloxedEGFP;NaV1.8Cre)胚胎(胚胎发育第18.5天,雌雄不限)中收获细胞,经荧光激活细胞分选(Fluorescence-Activated Cell Sorting,FACS)分离得到Ret阳性NaV1.8细胞,以此生成基因表达数据。该数据集可作为时间节点,用于比较分析Ret阳性伤害性感受器(nociceptor)在不同发育时间点的基因表达发生模式。研究人员从胚胎发育第18.5天的RetFloxedEGFP:NaV1.8Cre小鼠的背根神经节中分离细胞,经FACS分选后提取总RNA,并使用伊卢米娜HiSeq 2500(Illumina Hiseq 2500)测序平台完成转录组测序。具体实验流程可参考GUDMAP网站上Jain发布的相关方案。
