Approximately two hundred thousand female and male PGCs were isolated by FACS, from stage 12-16 embryos derived from vasa-EGFP/vasa-EGFP; nos-Gal4/nos-Gal4 females mated with UAS-RFP/Y males. EGFP and red RFP double-positive cells and EGFP-positive cells were isolated as female and male PGCs, respectively. Total RNAs were extracted from the isolated PGCs, and subjected to RNA-sequencing.
