Neurofibrillary Tangles (NFTs) caused by Tau protein hyperphosphorylation have been identified as the strongest correlate of cognitive dysfunction in Alzheimer’s Disease (AD). Although Glycogen Synthase Kinase 3 Beta (GSK-3β) is a promising target for preventing Tau hyperphosphorylation, the challenges in achieving high inhibitor selectivity while ensuring Blood-Brain Barrier (BBB) penetration have limited clinical progress. Utilizing our in silico Agency system—which leverages Recursive Resonance Modeling (RRM) and Quantum Logic simulation—we designed UmbraKinase-33, an ATP-competitive lead compound.
