Toll-like receptor (TLR)-induced zinc toxicity has emerged as an important component of the macrophage antimicrobial response against intracellular bacterial pathogens. Our data showed that human macrophage subjected EC958, a representative strain of the globally-disseminated multidrug resistant ST131 clone, to zinc stress. We therefore used transposon-directed insertion site sequencing (TraDIS) to identify the complete set of UPEC genes conferring protection against zinc toxicity. We incubated ~1 million EC958 transposon mutants in 1 mM ZnSO4 in LB broth for 18h, then recovered surviving cells and isolated genomic DNA. We then performed TraDIS and data analysis to identify both zinc-susceptibility and zinc-tolerance genes. The data submitted in this project were Illumina sequence data from two samples treated with LB as control and two samples treated with LB + 1 mM Zn.
