In order to determine the degree that mtDNA methylation regulates mtDNA replication during tumorigenesis, we have characterized the patterns of mtDNA methylation using tumour models of glioblastoma and osteosarcoma that have different combinations of mtDNA genotype and copy number against a nuclear genome background at different stages of tumour progression. The findings provide novel insights into mtDNA methylation in the epigenetic regulation of mtDNA copy number in tumorigenesis.
