Deficiency of gut-enriched Lipase H promotes gut aging and reduces lifespan in Drosophila

Liph, a gut-enriched Lipase H encoding gene, shows decreased expression during gut aging in both fruit fly and mouse. However, whether such evolutionary conserved Liph plays a protective role in gut aging remains unknown. In this study, RNA-seq was performed on gut tissue from fly model to identify changes in gene expression produced in whole guts after knocking down CG6295, the Drosophila ortholog of the mammalian Liph. In addition, we performed RNA-seq analysis on rat intestinal epithelial IEC-6 cells with Liph knockdown to investigate the role of Liph in mammalian intestinal aging.Liph（肠道富集型脂肪酶H编码基因）在果蝇与小鼠的肠道衰老过程中均呈现表达量下调。然而，这类进化保守的Liph是否在肠道衰老中发挥保护作用，目前仍未明确。本研究以果蝇模型的肠道组织为材料开展RNA测序（RNA-seq），以鉴定敲低哺乳动物Liph的果蝇同源基因CG6295后，整个肠道的基因表达变化情况。此外，本研究还对敲低Liph的大鼠肠上皮IEC-6细胞开展RNA测序分析，以探究Liph在哺乳动物肠道衰老中的作用。
