CRISPR knockout (CRISPR KO) screens on the PC cell line

To quantitively evaluate MetroSCREEN's performance in predicting MRs, we performed CRISPR knockout (CRISPR KO) screens on the PC cell line, targeting four well-known MRs: CTNNB159, ATF460, MYC61, and JUN62. Overall design: CRISPR knockout (CRISPR KO) screens on the PC cell line, targeting four well-known MRs: CTNNB, ATF, MYC, and JUN. After lentiviral library infection and puromycin selection, the sequences encoding the sgRNAs were PCR-amplified from the transduced cells and quantified by high-throughput sequencing. We conducted the screen with 10x Genomics droplet-based 3' scRNA-seq and direct sgRNA capture.
