Super-resolution STORM and STED microscopy data and derived image-analysis outputs for spectrin-dependent organization of sAHP-associated ion channels

This dataset contains super-resolution microscopy data and analysis outputs used to study the role of spectrin in the spatial organization of channels associated with the slow afterhyperpolarization (sAHP). The STORM component contains images and analysis outputs from primary rat hippocampal neurons immunolabelled for Spectrin Beta II (SPTBN1), IK (KCa3.1), RyR2, Actinin1, Actinin2, CaV1.3, Nav1.6, and MAP2. We used these data to evaluate channel distribution, nearest-neighbour relationships, and periodicity in somatic and validate in neuritic compartments. The STED component contains images and analysis outputs from rat brain sections labelled for Spectrin Alpha II (SPTAN1), IK (KCa3.1), and MAP2 under vehicle, maitotoxin, and calpeptin-plus-maitotoxin treatment conditions. We used these images to assess spectrin-dependent changes in cluster organization in tissue. The dataset includes raw or near-raw image products, renamed working images, deconvolved images, manually selected single-frame images, binary masks, overlap images, nearest-neighbour distance tables, non-negative matrix factorization outputs, autocorrelation and model-fitting outputs, statistical summaries. We also include MATLAB workspaces and scripts associated with analysis. We performed the analysis using Fiji/ImageJ, Huygens Professional, and MATLAB.
