We report the application of next generation sequencing of blood platelets in an early-early melanoma mice model for screening early cancer biomarkers. By inoculating C57BL/6 mice with different numbers of B16F10 cells, we establish an early-early melanoma model. We find that mice inoculated with lower number of B16F10 cells develop tumors at a later time. Therefore the blood samples collected at day 14 post-injection before the formation of visible tumors are early-early cancer samples. The mRNA profiles of platelets from early-early group are distinct from those of control group and large tumor group. However, the mRNA profiles of PBMCs from early-early group are not so distinct from those of control group and large tumor group. This study provides a proof of concept strategy for future cancer biomarker screening and cancer immunology research.
